No, you cannot. Not unless cheats are on or something.

You can do it with devmode, which can be very helpful for testing purposes, but it can't be done in normal gameplay.

Oh so no SA2 final story styled levels. Ok what is the change sking command in devmode?

I took this from Sonic Heroes(without permission, sorry =( ): http://www.4shared.com/file/j9F4lZMT/EasyChange.html
just add it in the game and press 1 for sonic, 2 for tails and 3 for knux.

Just erase the levels that you don't want to force the characters in the MAINCFG lump, or add a level that you want to force the characters, if the level isn't there.
